Longing to roam free in the vast landscapes of Mother Africa, Marty, the bored and dejected zebra of the famous Central Park Zoo, escapes his prison on the night of his tenth birthday celebration.
However, after a botched rescue attempt by Marty's companions--Alex, the content lion; Melman, the skittish giraffe, and Gloria, the resolute hippo--the friends will find themselves crated up and shipped off to a remote wildlife preserve, only to end up on the sandy shores of exotic Madagascar. The four, along with the penguins and two chimpanzees named Mason and Phil , find themselves at Grand Central Station, where they are quickly sedated via a tranquilizer gun when Alex's attempt to communicate with humans is mistaken for aggression.
The zoo, under pressure from anti-captivity activists, is forced to ship the escaped animals by sea to a Kenyan wildlife preserve. During their travels, the penguins escape from their enclosure and take over the ship, intent on taking it to Antarctica. Their antics on the bridge cause the crates containing Alex, Marty, Melman, and Gloria to fall overboard and wash ashore on Madagascar.
The animals are soon able to regroup, initially believing themselves to be at the San Diego Zoo. Alex blames Marty for their predicament and attempts to signal for help to get back to civilization. Marty, on the other hand, finds the wild to be exactly what he was looking for, with Gloria and Melman soon joining him in enjoying the island.
Alex eventually comes around, but, deprived of the raw steaks he was provided with at the zoo, his prey drive begins to show as hunger kicks in. The group is accepted by the lemurs, though King Julien's adviser, Maurice , cautions them about Alex's predatory nature. When the others find him gone, they head for Grand Central Station via the Subway to try to get him back.
Arriving at Grand Central, they scare away most of the passengers except for one determined old lady who assaults Alex by bashing his head and his groin with her handbag , and catch Marty. Then they manage to wreck the information desk before being shot with tranquilizer darts, along with the penguins, Mason and Phil two chimpanzees who also escaped.
Anti-zoo campaigners use the incident as evidence that animals should not be locked up. Because of this, all the animals soon find themselves in wooden crates aboard a cargo ship to Kenya.
The penguins succeed in taking over the ship and changing its course, following their initial plan of making their way to Antarctica. Alex is soon washed ashore on a tropical beach. All night he is alone, but in the morning, finds Melman stuck in his crate.
Gloria and Marty soon wash ashore on the same beach. They are actually in the wild, on the island of Madagascar. Alex blames Marty for deliberately ruining the group's lives at the zoo and divides the island between them, forcing Gloria and Melman to help him build a Statue of Liberty lookalike.
His plan is to ignite the beacon, therefore, making it easy for a boat which Alex believes is coming to rescue them to see where they are. However, Melman inadvertently sets the whole statue on fire and destroys it in a Planet of the Apes parody. Gloria and Melman then decide to join Marty who is living comfortably on his side of the island , and Alex soon realizes that he has been unfair and selfish to his friend.
The exotic animals on Madagascar are descended from ancestors who rafted to the island 60 million years ago — just like in the movie. Due to the continuing debate about whether or not the fifth installment in the franchise should be a reboot, a sequel, a remake, or a live-action film, Shrek 5 was almost canceled entirely.
